Early stages of development of terrestrial orchids: the effect of saccharides and phytohormones Rané fáze vývoje semenáčků teretsrických orchidejí: vliv sacharidů a fytohormonů Jan Ponert Úvod Mykorhiza je rozšířená u více než 90 % vyšších rostlin. Pro její studium však nelze využít klasický modelový organismus Arabidopsis thaliana, který mykorhizu nevytváří. Studium mykorhizních asociací tak dnes probíhá na různých organismech a modelový druh dosud zvolen nebyl. Jednou z rostlinných skupin, které by mohly být pro výzkum mykorhizních asociací velice vhodné, jsou orchideje (čeleď vstavačovité - Orchidaceae). Prakticky všechny druhy jsou ve svých raných vývojových fázích obligátně závislé na mykorhize. Semena obsahují jen velmi málo zásobních látek, a výživu mladých semenáčků tak musí zajistit mykorhizní houba. V pozdějších fázích vývoje většina orchidejí počíná fytosyntetizovat, mění svůj způsob výživy na částečnou mykotrofii a některé druhy mohou být v dospělosti zcela nezávislé na mykorhize. Jiné druhy však schopnost fotosyntézy ztratily a celý svůj vývojový cyklus se vyživují výhradně mykotrofií. Orchideje tak nabízejí možnost srovnání různých strategií mezi blízce příbuznými druhy, ale také mezi různými vývojovými fázemi jediného druhu. Early stages of development of terrestrial orchids: the effect of saccharides and phytohormones Rané fáze vývoje semenáčků teretsrických orchidejí: vliv sacharidů a fytohormonů Jan Ponert Abstract Orchids have very small seeds with only limited contents of reserves. Therefore, in early stages of their development most of them fully depend on nutrition provided by mycorrhizal fungi. Nearly all orchids remain partial mycotrophs during whole life, even after a start of their own photosynthesis. These characteristics make orchids an ideal model for studies of mycorrhizal associations and bring us good opportunity to study different feeding strategies in the single species. This work targets on early developmental stages of orchids and by studying carbohydrate status of both plant and fungus and their sensitivity to different types of phytohormons estimates potential of these signalling compounds to control plant-fungus coordination. Firstly, the capability of selected orchids to utilize different carbohydrates have been tested. Results confirmed their ability to metabolize sucrose, fructose, maltose, raffinose and trehalose.
