bioremediation, aromatic hydrocarbons, ring-hydroxylating dioxygenasesLokalita bývalého vojenského letiště Hradčany je jedním z míst s vysokou koncentrací organických polutantů v půdě v rámci České republiky. Hlavním nástrojem sanací probíhajících v této lokalitě v současné době je bioremediace. Jsou při ní využívány organismy přítomné v kontaminované půdě disponující degradačními metabolickými drahami pro dekontaminaci lokality in situ. Na rozmanitosti a hojnosti těchto drah, resp. na specifitě a aktivitě enzymů podílejících se na těchto drahách závisí účinnost bioremediace. Proto je hlavním cílem této práce analýza diversity bakterií v půdách kontaminovaných leteckým palivem na základě katabolických genů kódujících toluen/bifenylové dioxygenasy Rieskeho typu. Z půdní DNA byly selektivně získány sekvence kódující tyto geny pomocí hybridizace s oligonukleotidy vázanými na magnetické mikrokuličky. podjednotky těchto dioxygenas byly amplifikovány, analyzovány klonováním, restrikční analýzou a sekvenovány. Evoluční historie byla odvozena pomocí metod neighbor-joining a maximum likelihood. Diversita katabolických genů v půdě HRB z kontaminované a sanované lokality byla porovnána s diversitou ve stejné půdě použité v mesokosmu se simulovanou fytoremediací. The former military air-base Hradcany is among the most contaminated with organic pollutants localities in Czech Republic. Main cleanup strategy in the area is the bioremediation taking advantage on the natural potential of the autochthonous soil microorganisms to evolve catabolic pathways for in situ degradation of the pollutant. The diversity and abundance of the pathways, as well as the specificity and activity of the encoded enzymes are priority biotic factors determining the bioremediation efficiency. Main task of this work was to analyze the bacterial diversity in jet fuel contaminated soils based on key catabolic genes encoding the Rieske non-haem iron dioxygenases of the toluene/ biphenyl oxygenase branch. High molecular soil DNA was extracted and the sequences encoding catabolic genes were selectively enriched by hybridization to biotinylated oligonucleotides on magnetic microbeads with covalently bound streptavidin. Fragments of the genes for the -subunits of Rieske non-haem iron oxygenases were amplified and analyzed by restriction analysis, cloning and sequencing.
