ubiquitin ligases, protein degradation, FBXO38, centromeres, spermatogenesisUbikvitin ligázy jsou zodpovědné za specifické rozpoznání proteinů určených k odstranění pomocí proteazomu. Cílem našeho projektu byla molekulární a funkční charakterizace ubikvitin ligázy SCFFBXO38 . Podobně jako u mnoha jiných není její biologická funkce dosud podrobně objasněna, přestože se jedná o jedinou ubikvitin ligázu, jejíž mutace vedou k rozvoji distální formy svalové atrofie. V první části našeho projektu jsme identifikovali nové substráty této ubikvitin ligázy, a to jaderné proteiny ZXDA a ZXDB, jejichž funkce není dosud popsána. Pomocí genetických a biochemických metod jsme prokázali, že proteiny ZXDA/B jsou pozitivními regulátory integrity centromerického chromatinu, a že experimentální inaktivace ubikvitin ligázy SCFFBXO38 vede ke stabilizaci proteinů CENP-A a CENP-B v oblasti centromer, a to v závislosti na ZXDA/B. Ve druhé části projektu jsme se zaměřili na analýzu myšího modelu s delecí genu Fbxo38. Prokázali jsme, že ztráta Fbxo38 vede k poruchám růstu, které postihují různé orgány včetně samčí reprodukční soustavy. Podrobnou histologickou analýzou jsme odhalili patologické změny v semenotvorných kanálcích doprovázené nižší produkcí spermií a sníženou fertilitou. Ubiquitin ligases are responsible for the specific recognition of proteins targeted for proteasome-dependent degradation. This project focused on the molecular and functional characterization of the SCFFBXO38 ubiquitin ligase. As with many others, its biological function has not yet been elucidated in detail, although it is the only ubiquitin ligase whose mutations lead to the onset of a distal form of muscle atrophy. In the first part of our project, we identified new substrates for this ubiquitin ligase, the nuclear proteins ZXDA and ZXDB, with insufficiently characterized functions. Using genetic and biochemical methods, we have shown that ZXDA/B proteins act as positive regulators of centromeric chromatin integrity and that experimental inactivation of the SCFFBXO38 ubiquitin ligase resulted in a ZXDA/B-dependent stabilization of CENP-A and CENP-B proteins in the centromeric regions. In the second part of the project, we focused on analyzing the mouse model deficient in the Fbxo38 gene. We demonstrated that loss of Fbxo38 leads to growth retardation affecting various organs, including the male reproductive system.
