HBV, chronic hepatitis B, treatment, STINGChronická hepatitida B (CHB) je infekční onemocnění způsobené hepatotropním virem hepatitidy B (HBV). CHB celosvětově trpí miliony lidí, ale možnosti léčby tohoto onemocnění jsou stále limitované a k úplně eliminaci viru dochází jen málokdy. V současnosti se začínají zkoumat imunomodulační přístupy v léčbě CHB. V této práci jsme se zaměřili na anti-HBV potenciál dráhy přirozené imunity rozpoznávající DNA, tzv. dráhy syntázy cyklického GMP-AMP-stimulátoru genů pro interferony (cGAS-STING). Aktivace této dráhy vede k sekreci prozánětlivých cytokinů. V naší laboratoři jsme otestovali připravené syntetické aktivátory proteinu STING založené na cyklických dinukleotidech (CDN) a z nich vytvořené lipofilní prolátky v reportérových buněčných liniích a jednojaderných buňkách periferní krve (PBMC). Vybrali jsme kandidátní molekuly, které aktivovaly dráhu cGAS-STING lépe než přirozený eukaryotický aktivátor proteinu STING. Identifikovali jsme, že 3′,3′-c-di(2′-fluoro,2′-deoxyAMP) lépe aktivoval dráhu cGAS-STING napříč pěti isoformami proteinu STING nejvíce zastoupenými v lidské populaci včetně těch, které jsou špatně aktivovány 3′,3′-CDN. Symetrická bis-pivaloyloxymethylová prolátka připravená z 3′,3′-c- di(2′-fluoro,2′-deoxyAMP) dále zlepšila vlastnosti CDN tím, že usnadnila vstup CDN do buňky. Ukázali jsme,...
Chronic hepatitis B (CHB) is an infectious disease caused by the hepatotropic hepatitis B virus (HBV). CHB affects millions of people worldwide, but the treatment options are still limited, and virus elimination is rarely achieved. The immunomodulatory approaches towards CHB treatment are recently being investigated. In this thesis, we focused on the anti-HBV potential of the cyclic- GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) DNA-sensing innate immunity pathway, which upon activation leads to the secretion of proinflammatory cytokines. We screened synthetic cyclic dinucleotide (CDN)-based STING agonists and their lipophilic prodrugs prepared in our laboratory in reporter cell lines and peripheral blood mononuclear cells (PBMC). We selected candidate compounds with superior cGAS-STING pathway activation properties compared to the natural eukaryotic STING agonist. We identified 3′,3′-c-di(2′-fluoro,2′- deoxyAMP) to have improved cGAS-STING pathway activation properties over the five most abundant STING isoforms found in the humans, including the variants that do not respond well to 3′,3′-CDNs.
