Celá řada miRNA se podílí na patogenezi kardiovaskulárních onemocnění, což je asociováno se změnou jejich genové exprese. Cílem této práce bylo zjistit, jak se liší genová exprese miRNA mezi jednotlivými biologickými zdroji - plnou periferní žilní krví (plná PK), plazmou a plazmatickými exozómy. U celého souboru žen byly expresní hladiny miRNA nejvyšší v plné PK v porovnání s plazmou či plazmatickými exozómy. U celého souboru žen byly expresní hladiny miRNA vyšší v plazmě v porovnání s plazmatickými exozómy, kromě miR-126-3p, kde byla detekována vyšší hladina v plazmatických exozómech ve srovnání s plazmou. V této práci byly také porovnány expresní hladiny kardiovaskulárních miRNA mezi ženami s anamnézou gestačního diabetu mellitu (GDM) a ženami s anamnézou fyziologické gravidity (FG) v období 3-11 let postpartum v plné PK, plazmě a plazmatických exozómech. V plné PK bylo 12 z 29 testovaných miRNA up-regulováno u žen s anamnézou GDM. V plazmatických exozómech byla up-regulována miR-181a-5p a v plazmě miR-499a-5p u žen s anamnézou GDM. Změny v plné PK zřejmě odrážejí komplexní systémovou reakci na změny, které nastaly v důsledku klinické manifestace GDM. U žen s aberantním epigenetickým profilem...
MicroRNA (miRNA) are short non-coding RNA molecules that regulate gene expression at the post-transcriptional level. Many miRNAs are involved in the pathogenesis of cardiovascular diseases, which is associated with altered gene expression. This work compares miRNA gene expression profiles among various biological sources - whole peripheral venous blood (whole PB), plasma and plasma exosomes. For all tested groups combined, the expression levels of miRNA were maximal in whole PB and lowered in plasma and plasma exosomes, and the expression levels of miRNA were higher in plasma than in plasma exosomes, except miR-126-3p, which had a higher level detected in plasma exosomes compared to plasma. This work also compares expression levels of cardiovascular miRNA between women with anamnesis of gestational diabetes mellitus (GDM) and physiological gravidity 3-11 years postpartum in whole PB, plasma and plasma exosomes. In whole PB, 12 of 29 tested miRNAs were up-regulated in women with prior exposure to GDM. MiR-181a-5p was up-regulated in plasma exosomes and miR-499a-5p in plasma in women with prior exposure to GDM.
