Vandetanib, nanoparticles, apoferritin, liposomes, targeted medicine4 Abstrakt V rámci této diplomové práce bylo studováno protinádorové léčivo vandetanib. Vandetanib je tyrosin kinasový inhibitor působící zejména na receptory vaskulárních endoteliálních růstových faktorů (VEGFR), epidermálních růstových faktorů (EGFR) a RET protoonkogeny (REarranged during Transfection). Používá se primárně k léčbě pokročilých stádií nádorů štítné žlázy. Bohužel použití vandetanibu k léčbě je značně limitováno pro jeho toxicitu a zejména kardiotoxicitu (dochází k prodloužení QT intervalu). Jeden ze způsobů, jak tuto toxicitu minimalizovat, je využití nanočástice k transportu inhibitoru do místa působení. Jako transportní nanočástice byl použit apoferritin a liposomy. Cílem předkládané diplomové práce bylo sledování stability nanočástice apoferritinu s molekulami inhibitoru vandetanibu (ApoVan) a sledování vlivu pH na uvolňování inhibitoru z ApoVan formy. Provedenými experimenty bylo zjištěno, že ApoVan komplex je poměrně stabilní po skladování při 4 řC a - 20 řC po dobu až 8 týdnů. Bohužel při sledování vlivu pH na uvolňování inhibitoru z ApoVan se zjistilo, že se vandetanib postupně uvolňuje nejen do kyselého pH (6,5), ale i do fyziologického pH (7,4) a způsob, jakým je ApoVan připraven, na to nemá žádný vliv. 5 Abstract In this thesis the anticancer drug vandetanib was studied. Vandetanib is a tyrosine kinase inhibitor affecting signalling of vascular endothelial growth factor receptor (VEGFR), epidermal growth factor receptor (EGFR) or RET protooncogene (REarranged during Transfection). It is primarily used for the treatment of advanced tumors of the thyroid gland. Unfortunately, the usage of vandetanib in the cancer treatment is significantly limited by its toxicity and cardiotoxicity (one of the adverse effects is connected with long QT interval). One way, how to minimize these side effects, is binding a drug into a suitable transporter. Apoferritin and liposomes were used as a transport nanoparticles in this study. The aim of this thesis was to study the stability of the complex of nanoparticle apoferritin with vandetanib molecules (ApoVan) and to study the effect of pH on the release of inhibitor from the ApoVan form. Experiments have shown that ApoVan complex is relatively stable after its storage at 4 řC and - 20 řC for up to 8 weeks.
