boron doped diamond electrode, dehydration, glassy carbon electrode, phytosterols, oxidation, reduction, voltammetryCílem této diplomové práce bylo vyvinout elektrochemickou metodu pro tři fytosteroly, a to stigmasterol, β-sitosterol a ergosterol za pomoci voltametrických technik na elektrodách ze skelného uhlíku a bórem dopovaného diamantu. Práce navazuje na bakalářskou práci [1], kde se ukázalo, že k elektrochemické oxidaci fytosterolů je potřeba kyseliny chloristé v acetonitrilu. Při použití této kyseliny dochází k dehydratační reakci a produkty této dehydratace jsou elektrochemicky oxidovány. Kalibrační závislosti byly naměřeny pro všechny fytosteroly v prostředí s obsahem vody 0,43 %. Stálost signálu byla sledována při zvyšujícím se obsahu deionizované vody. Poté byly změřeny kalibrační závislosti ve zvýšeném obsahu vody. Pokud budeme chtít použít separační techniky typu HPLC, musíme nejprve zjistit, jak se budou fytosteroly chovat v tomto prostředí, a proto se zkoumala stálost signálu na obsahu pufrů. Použitými pufry byly fosfátový a acetátový pufr. Posléze byla naměřena koncentrační závislost v prostředí s vyšším obsahem fosfátového pufru. Dále byla zkoumána možnost detekce dvou fytosterolů zároveň při jednom měření. The aim of this diploma thesis was to develop an electrochemical method for three phytosterols, namely stigmasterol β-sitosterol and ergosterol using voltammetry techniques on glassy carbon electrode and boron doped diamond electrode. The work builds on previous results, where it was shown that the electrochemical oxidation of phytosterols requires perchloric acid in acetonitrile. When this acid is used, a dehydration reaction occurs. Calibration dependences were measured here for all phytosterols in an environment with a water content of 0.43%. The stability of the signal with increasing distilled water content was further investigated. Then the calibration dependences in the increased water content were measured again. If we want to use HPLC-type separation techniques, we must first find out how the phytosterols will behave in this environment, and therefore the signal stability on the buffer content was investigated. The buffers used were phosphate and acetate buffer. Subsequently, the concentration dependence was measured in an environment with a higher content of phosphate buffer. Furthermore, the possibility of detecting two phytosterols at once was investigated.
