5 Abstrakt Acylceramidy, podskupina ceramidov s ultradlhými reťazcami sú esenciálnou zložkou extracelulárnej lipidovej matrix v najvrchnejšej vrstve kože, stratum corneum. Majú nezastupiteľnú funkciu v prežití cicavcov na súši. Hlbšie pochopenie ich funkcie vo fyziológii a patofyziológii kože a ich terapeutického použitia je obmedzené ich limitovanou dostupnosťou. Analýza povrchových lipidov osla (E. asinus) ukázala, že v týchto lipidoch sa nachádza až 56 % nerozvetvených ω-laktónov (equolidov), z ktorých 51,2 % tvorí nenasýtený dotriakontanolid a 41,3 % nenasýtený triakontanolid. Tieto laktóny sa svojou dĺžkou reťazca zhodujú s najbežnejšou dĺžkou acylového reťazca acylceramidov (30 a 32 atómov uhlíka) a mohli by teda nájsť uplatnenie pri ich totálnej syntéze. Cieľom tejto práce bolo izolovať nenasýtený ω-laktón s 32 uhlíkovým skeletom (dotriakontanolid) zo zmesi povrchových lipidov osla, následne ho hydrogenovať a previesť na vhodný prekurzor (sukcinimidyl ester) a nájsť tak čo najjednoduchšiu syntetickú cestu jeho premeny na acylceramidy. Vyskúšali sme mnoho syntetických ciest. Od priamej aminolýzy laktónu, cez reakciu s N- hydroxysukcinimidom v rôznych reakčných podmienkach po otvorenie laktónu na draselnú soľ ω-hydroxykyseliny, nasledované reakciou s disukcinimidyl karbonátom. 6 Abstract Acylceramides, subgroup of ceramides with ultralong chains, are essential component of extracellular lipid matrix in the uppermost skin layer, stratum corneum. They have crucial role in mammalian survival on dry land. Deeper understanding of their function in physiology of pathophysiology of the skin and their therapeutic potential are hampered by their limited availability. Analysis of the skin surface lipids of the ass (E. asinus) has shown, that these lipids contain up to 56% of unbranched ω-lactones (equolides), from which 51.2% is mono- unsaturated dotriacontanolide and 41.3% is mono-unsaturated triacontanolide. Carbon chain length of these lactones match the most common length of carbon chain in acylceramides (30 and 32 carbon atoms) therefore they could be used in their total synthesis. Aim of this thesis was to isolate mono-unsaturated ω-lactone with 32 carbon chain (dotriacontanolide) from the mixture of donkey skin surface lipids, followed by hydrogenation and transformation to the suitable precursor (succinmidyl ester) in order to find the easiest synthetic path in its conversion to acylceramides.
