Characterization of DNA binding of CSL transcription factors in fission yeast

dc.description.abstractProteiny Cbf11 a Cbf12, patřící do rodiny transkripčních faktorů CSL, jsou u poltivé kvasinky Schizosaccharomyces pombe zapojeny v široké škále buněčných procesů - mj. regulují buněčnou adhezi a podílejí se na udržování integrity genomu. V rámci celého genomu jsme již dříve identifikovali cílové lokusy, kam se proteiny CSL váží in vivo. Mnoho z cílových míst neobsahuje konsenzuální CSL-vazebný element. Patrně existují různé režimy vazby proteinů Cbf11/12 na DNA a doposud nebylo známo, s jakou biologickou funkcí je způsob vazby na DNA spojen. Pro účely studia DNA-vazebných módů proteinů CSL jsme v rámci tohoto projektu pracovali na zavedení DNA-vazebné mutace (DBM), která způsobuje ztrátu schopnosti proteinů CSL se vázat na kanonický motiv in vitro, do chromozomálního lokusu genů cbf11 a cbf12. S použitím "ura4 selekčního systému" jsme úspěšně zkonstruovali kmeny Cbf12-TAP a Cbf12DBM-TAP, tj. kmeny bez/s DBM v otevřeném čtecím rámci Cbf12, ve kterých je navíc protein Cbf12 C-terminálně značen TAP-tagem a obsahuje intaktní oblast 3'UTR. Nově jsme v naší laboratoři zavedli CRISPR/Cas9 systém, s využitím kterého se nám podařilo připravit kmen Cbf11-TAP. S konstrukcí kmene Cbf11DBM-TAP jsme prozatím neuspěli, nicméně v rámci práce diskutujeme možné optimalizace konstrukčního postupu, které by mohly vést...cs_CZ
dc.description.abstractCbf11 and Cbf12 proteins, the members of the CSL transcription factors family, are involved in a wide range of cellular processes in the fission yeast Schizosaccharomyces pombe - among other things they regulate cell adhesion and they have also been implicated in maintenance of genome integrity. At the level of the whole genome we previously identified target loci bound by CSL proteins in vivo. Many of them do not contain any consensus CSL-binding element. There are probably different DNA binding modes of the Cbf11/12 proteins and it has not been known what specific biological function is associated with the particular way of DNA binding. For the purpose of studying CSL DNA binding modes we have worked in this project on the implementation of the DNA binding mutation (DBM), which prevents direct DNA binding of CSL proteins to canonical motif in vitro, into the chromosomal locus of the cbf11 and cbf12 genes. Using the "ura4 selection system" we have successfully constructed the scar-less Cbf12-TAP and Cbf12DBM-TAP knock-ins, i.e. the strains without/with DBM in the open reading frame of Cbf12 where Cbf12 is C- terminally TAP-tagged and contains the intact 3'UTR.
