dc.description.abstract | Turnerův syndrom (TS) je genetické onemocnění žen s celosvětovou frekvencí výskytu 1:2 000. Jeho příčinou nemusí být pouze monozómie chromozómu X (45,X), nýbrž i mozaikový karyotyp (45,X/46,XX; 45,X/46,XY; 45,X/47,XXX), izochromozóm X (46,X,i(Xq)), ring chromozóm X (45,X/46,X,r(X)) nebo další strukturní aberace chromozómu X (delece a translokace). U pacientek s TS (s výjimkou pacientek s ring chromozómem) není diagnostikována mentální retardace. TS úzce souvisí s regulací exprese genové dávky, především s procesem inaktivace chromozómu X. Geny unikající procesu inaktivace chromozómu X, jsou u ženy karyotypu 45,X exprimovány v nesprávné, poloviční dávce ve srovnání se zdravou ženou. Příkladem takového genu je SHOX, na který jsem se soustředila ve své bakalářské práci. Haploinsuficience genu SHOX způsobuje majoritní fenotypový projev TS, kterým je malá postava a kostní abnormality. Výzkum dalších genů, odpovědných za TS, komplikuje skutečnost, že neexistuje vhodný modelový organizmus, na kterém by bylo možné TS studovat a výsledky extrapolovat na člověka. U myších samic se liší nejen samotný proces inaktivace chromozómu X, ale i fenotypové projevy s karyotypem 39,X jsou daleko mírnější než u žen s karyotypem 45,X, což lze vysvětlit skutečností, že u myši uniká procesu inaktivace chromozómu X pouze... | cs_CZ |
dc.description.abstract | Turner syndrome (TS) is a genetic anomaly occurring in women with worldwide frequency 1:2,000. Turner syndrome's possible causes include X chromosome monosomy, mosaic karyotype (45,X/46,XX; 45,X/46,XY; 45,X/47,XXX), X isochromosome (46,X,i(Xq)), ring X chromosome (45,X/46,X,r(X)), and other X chromosome aberrations (deletion and translocation). Patients with Turner syndrome (with the exception of ring chromosome abnormalities) aren't diagnosed with mental retardation. Turner syndrome is closely related to the regulation of gene expression level, particularly with X chromosome inactivation. Genes escaping the X chromosome inactivation process in 45,X women are expressed in half the dose of a healthy woman. An example of such a gene is SHOX, which I decided to focuse on in this thesis. SHOX gene haploinsufficiency causes major Turner syndrome phenotype manifesting of short body and bone abnormalities. The research of other genes with possible roles in Turner syndrome is complicated by the absence of adequate model organism, which could be used for TS study with possibility to extrapolate the results to humans.
