New markers of colon cancer evolution in ulcerative colitis

dc.description.abstractBackground: Long-standing ulcerative colitis (UC) has an increased risk of evolving into colorectal cancer (CRC) and upregulated expression of cyclooxygenase 2 (COX-2), inducible nitric oxide synthase (iNOS), survivin, telomerase catalytic subunit (TERT), integrin-linked kinase (ILK) and transcription factors c- MYB and TCF-4, has been implicated in the development and progression of sporadic colorectal cancer. Nevertheless much less is known about their role in the process of UC-associated colon carcinogenesis. Methods: We analyzed the gene expression of these markers during the transition of colonic mucosa from chronic inflammation to epithelial neoplasia in biopsies of UC patients using quantitative real-time polymerase chain reaction and immunohistochemistry, and compared the expression profiles of this gene panel in samples of patients with CRC and in human tumor xenografts of SW620 malignant colonic cells. Additionally, we determined the expression of these genes in mouse models of sporadic and colitis-associated CRC in A/J and ICR mouse strains using quantitative RT-PCR and laser microdissection. dc.description.abstractSOUHRN Úvod: Dlouhotrvající ulcerozní kolitida (UC) je spojena se zvýšeným rizikem vývoje kolorektálního karcinomu (CRC) a zvýsená exprese cyklooxygenázy 2 (COX-2), inducibilní NO syntázy (iNOS), survivinu, telomerázové katalytické podjednotky (TERT), integrinové kinázy (ILK) a transkripčních faktorů c-MYB a TCF-4 byla prokázána během sporadické kolorektální tumorigeneze. Nicméně změny v expresi těchto proliferačních a antiapoptotických genů během neoplastické transformace u ulcerozní kolitidy nejsou dostatečně popsány. Metody: Analyzovali jsem genovou expresi těchto markerů během přechodu epitelu tlustého střeva od chronickým zánětem iritované mukozy k epiteliální neoplasii v biopsiích pacientů s UC pomocí kvantitativní RT-PCR a imunohistochemie a porovnali jsme změny genové exprese studovaných markerů také ve vzorcích získaných od pacientů operovaných pro sporadický kolorektální karcinom a v xenograftových vzorcích odvozených od nádorové linie SW620. Dále jsme studovali expresi mRNA těchto genů také s využitím laserové mikrodisekce u myšího modelu s kolitidou asociované (kmen ICR) i sporadické kolorektální karcinogeneze (kmen A/J). Výsledky: Hladiny mRNA pro survivin, c-MYB, COX-2, iNOS a TCF-4 byly signifikantně zvýšeny během neoplastické transformace epitelu u pacientů s ulcerozní kolitidou, naopak TERT...cs_CZ
