dc.description.abstractAbstraсt Mutations in core spliceosomal proteins, including PRPF8, are associated with autosomal dominant retinitis pigmentosa (adRP), a progressive retinal degenerative disease. Despite the ubiquitous expression of splicing factors, the molecular mechanisms underlying retina-specific pathology remain poorly understood. In this study, we investigated the effects of the PRPF8-Y2334N mutation on RNA splicing and gene expression using human induced pluripotent stem cells-derived retinal pigment epithelium (hiPSC-derived RPE) cells. The mutation did not affect hiPSC-derived RPE differentiation or morphology, as confirmed by marker expression and deconvolution of RNA sequencing (RNA-seq) data. However, RNA-seq revealed widespread transcriptomic alterations, including differential gene expression and extensive changes in alternative splicing. Splicing analysis revealed that exon skipping and intron retention were among the most prominent alterations. Notably, intron retention affected genes encoding splicing regulators, suggesting disruption of autoregulatory feedback mechanisms within the splicing machinery. dc.description.abstractMutace v základních proteinech sestřihového komplexu, včetně PRPF8, jsou spojeny s autozomálně dominantní retinitis pigmentosa (adRP), progresivním degenerativním onemocněním sítnice. Navzdory ubikvitární expresi sestřihových faktorů zůstávají molekulární mechanismy, které stojí za tkáňově specifickou patologií sítnice, nedostatečně objasněny. V této studii jsme zkoumali vliv mutace PRPF8-Y2334N na sestřih RNA a genovou expresi za použití buněk retinálního pigmentového epitelu odvozených z lidských indukovaných pluripotentních kmenových buněk (hiPSC-derived RPE). Mutace neovlivnila diferenciaci ani morfologii hiPSC-derived RPE buněk, což bylo potvrzeno expresí markerů a dekonvolucí dat ze sekvenování RNA (RNA-seq). RNA-seq analýza však odhalila rozsáhlé transkriptomické změny, včetně diferenciální genové exprese a výrazných změn v alternativním sestřihu. Analýza sestřihu ukázala, že mezi nejvýraznější změny patřilo vynechávání exonů a retence intronů. Retence intronů významně zasahovala geny kódující regulátory sestřihu, což naznačuje narušení autoregulačních zpětnovazebných mechanismů v rámci sestřihového aparátu.
