dc.description.abstractNatural killer cells (NK cells) play a key role in the first line of defense against tumor cells and cells infected by viruses. Particularly interesting is the unique mechanism of NK cell activation known as missing-self recognition. It involves the detection of decreased expression of naturally occurring inhibitory ligands for NK cell receptors on the surface of tumor cells or virus-infected cells. An example of such ligand is the protein Clr-11, which is expressed on the surface of healthy cells in rats and interacts with the NK cell receptor NKR-P1B. Over the course of evolution, some viruses managed to develop mechanisms that allow them to evade the NK cell-mediated immune response. In relation to the receptor:ligand pair NKR-P1B:Clr-11, such an adaptation was found in the rat cytomegalovirus. This virus carries a gene encoding the RCTL protein, which is an ortholog of Clr-11. Upon infection of a healthy cell by this virus, the surface expression of Clr-11 is downregulated, which would trigger an NK cell immune response under normal conditions. However, expression of RCTL protein is simultaneously induced, and through the interaction of this protein with NKR-P1B, the inhibitory signal is maintained, thereby preventing NK cell activation. dc.description.abstractPřirozeně zabijácké buňky (NK buňky) hrají klíčovou roli v první linii obrany proti nádorovým a virem infikovaným buňkám. Zajímavý je unikátní způsob aktivace NK buněk označovaný jako "missing-self". Ten spočívá v detekci snížené exprese inhibičních ligandů pro receptory NK buněk na povrchu virem infikovaných či nádorových buněk. Příkladem takového ligandu je protein Clr-11, který je exprimován na povrchu zdravých buněk potkanů a interaguje s NK-buněčným receptorem NKR-P1B. V průběhu evoluce si některé druhy virů dokázaly vyvinout mechanismy, které jim umožňují vyhnout se imunitní reakci NK buněk. V souvislosti s receptor:ligandovým párem NKR-P1B:Clr-11 byla taková adaptace nalezena u potkaního cytomegaloviru. Ten ve svém genomu nese gen pro RCTL, orthologní protein k Clr-11. Po infekci zdravé buňky tímto virem dochází ke snížení exprese proteinu Clr-11 na jejím povrchu, což by za běžných okolností vedlo k NK-buněčné imunitní odpovědi. Současně se však spustí exprese proteinu RCTL, který interakcí s NKR-P1B zachová inhibiční signál a zabrání tak aktivaci NK buněk. Cílem této práce je rekombinantní produkce proteinu RCTL s N-glykosylací divokého typu v buněčné linii HEK293T a s uniformní glykosylací v buňkách Expi293F GnTI- pro další účely.
