Effect of frequently used drugs on growth, antimicrobial resistance and virulence of methicillin resistant Staphylococcus aureus

dc.description.abstractMeticilin-rezistentní Staphylococcus aureus (MRSA) je charakterizován výskytem dominantních klonálních linií, jejichž zastoupení se v průběhu času mění. S ohledem na variabilitu těchto linií jsme analyzovali soubor izolátů reprezentujících dominantní klonální linie MRSA v České republice. Studie zahrnovala izoláty linií převažujících po roce 2003 (ST225, ST22), komunitní kmen MRSA USA300 (ST8) a historické klonální linie (ST239(BC), ST239(CC), ST247) izolované v roce 1999. Po roce 2003 došlo k výraznému nárůstu prevalence infekcí způsobených MRSA, což je přičítáno specifickým adaptačním vlastnostem dominantního klonu ST225 (tzv. Rýnsko-Hessenský klon). V rámci studie jsme zkoumali, zda může přítomnost terapeutických koncentrací neantibiotických léčiv (kyselina acetylsalicylová, ibuprofen a paracetamol) v lidském těle a prostředí ovlivnit selekci dominantních klonů MRSA. Výsledky ukázaly, že i když tato léčiva nemají významný inhibiční účinek na testované izoláty a nelze je samostatně využít k terapii MRSA infekcí, může jejich přítomnost u některých izolátů (ST22 a ST225) způsobit významné zpomalení růstu. Dále bylo pozorováno zvýšení produkce biofilmu u izolátů ST239(BC), ST8 a ST225 v přítomnosti paracetamolu, přičemž aspirin tento efekt vyvolal u izolátů ST239(BC), ST247 a ST8. dc.description.abstractMethicillin-resistant Staphylococcus aureus (MRSA) is characterized by the presence of dominant clonal lineages, whose distribution changes over time. Given the variability of these lineages, we analyzed a set of isolates representing the dominant MRSA clonal lineages in the Czech Republic. The study included isolates from lineages prevailing after 2003 (ST225, ST22), the community-associated MRSA strain USA300 (ST8), and historical clonal lineages (ST239(BC), ST239(CC), ST247) isolated in 1999. Since 2003, a significant increase in the prevalence of MRSA infections has been observed, which is attributed to the specific adaptive properties of the dominant clone ST225 (also known as the Rhine-Hesse clone). As part of the study, we investigated whether the presence of subinhibitory concentrations of non- antibiotic drugs (acetylsalicylic acid, ibuprofen, and paracetamol) in the human body and environment could influence the dynamics of dominant MRSA clones. The results showed that these drugs cannot be used alone for the treatment of MRSA infections. However, they significantly slowed the growth of ST22 and ST225 isolates.
