Impact of mitochondrial complex I deficiency on the schizophrenia pathogenesis

dc.description.abstractSchizophrenia is a complex disorder and one of the most severe psychiatric conditions, yet its etiology remains poorly understood. It is associated with a wide range of pathophysiological processes, including mitochondrial dysfunction. Currently, clinical practice lacks a peripheral biological marker that could assist in the early detection and diagnosis of schizophrenia. One potential marker is dysfunction of mitochondrial complex I, which can be detected in both the brain and peripheral tissues of affected individuals. This thesis summarizes the current findings on complex I alterations in the context of mitochondrial dysfunction in schizophrenia and explores their potential diagnostic value. Changes in the expression of the NDUFV1, NDUFV2, and NDUFS1 subunits, as well as alterations in the enzymatic activity of complex I, are described as potential markers. The thesis links complex I dysfunction to increased oxidative stress in patients with schizophrenia, including the potential therapeutic use of antioxidants. In the final section, the inhibitory effect of antipsychotics on complex I and the possible clinical implications of these interactions are discussed. dc.description.abstractSchizofrenie patří k nejzávažnějším psychiatrickým onemocněním, její etiologie však není dostatečně objasněna. Pojí se se širokou škálou patofyziologických procesů, mezi které patří i mitochondriální dysfunkce. Současná klinická praxe postrádá periferní biologický marker, který by pomohl schizofrenii včasně zachytit a diagnostikovat. Jedním z možných markerů schizofrenie jsou poruchy mitochondriálního komplexu I detekovatelné v mozku i v periferních tkáních pacientů. Tato práce shrnuje dosavadní poznatky o poruchách komplexu I v kontextu mitochondriálních dysfunkcí u schizofreniků a o jejich možném diagnostickém využití. Jako potenciální marker jsou zde popsány změny exprese podjednotek NDUFV1, NDUFV2 a NDUFS1 a odlišnosti enzymatické aktivity komplexu I. Práce dává do souvislosti poruchy komplexu I se zvýšeným oxidačním stresem u pacientů se schizofrenií včetně možnosti terapeutického využití antioxidantů. V poslední části je diskutován inhibiční efekt antipsychotik na komplex I a možné klinické důsledky těchto interakcí.
