Establishment and characterisation of SP6A knock-out mutants as a tool to study tuber formation in cultivated potato

dc.description.abstractLilek brambor (Solanum tuberosum) je čtvrtá nejdůležitější zemědělská plodina. S růstem světové populace bude potřeba zajistit a udržet stabilní výnosy, které jsou nyní ohroženy postupující klimatickou změnou, a to například negativním zásahem do iniciace tvorby hlíz. Tuberizace je komplexní děj, zahrnující celou řadu regulačních faktorů, kdy změna každého z nich se může odrazit v úspěšnosti celého procesu. Mezi důležité tuberigenní faktory patří mobilní protein SP6A. Cílem práce bylo na základě charakterizace linií s knock-outovaným SP6A genem objasnit úlohu SP6A v procesu tuberizace u kulturního bramboru. Pro mutagenezi jsem použila metodu CRISPR/Cas9 se specificky navrženou gRNA tak, aby došlo k editaci všech SP6A alel této tetraploidní rostliny. Komponenty CRISPR/Cas9 jsem do rostlinného materiálu stabilně začlenila prostřednictvím Agrobacterium tumefaciens. Následná regenerace de novo nevedla k odvození ztrátového mutanta. Opakovaná regenerace de novo sice zvýšila efektivitu editace, nicméně jsem u dosud testovaných linií nezískala kompletního knock-out mutanta. U části odvozených linií byla zjištěna osminukleotidová delece v sekvenované oblasti sledovaného genu, nicméně kromě editovaných sekvencí zůstala v amplikonu i podstatná část needitovaná. dc.description.abstractPotato (Solanum tuberosum) is the fourth most important crop. As the world's population grows, there will be a need to ensure and maintain stable yields, which are now threatened by advancing climate change, for example by negative impact on tuber initiation. Tuberisation is a complex process governed by a number of regulatory factors, and changes in each of them can affect the success of the whole process. Important tuberigenic factors include the mobile protein SP6A. The aim of this study was to elucidate the role of SP6A in the tuberization process in cultivated potato by characterizing lines with SP6A gene knock-out. For mutagenesis, I used the CRISPR/Cas9 method with specifically designed gRNA so that all SP6A alleles of this tetraploid species have been edited. I stably incorporated the CRISPR/Cas9 components into the plant material via Agrobacterium tumefaciens. Subsequent de novo regeneration did not result in the derivation of a loss-of-function mutant. Although repeated de novo regeneration increased the editing efficiency, I did not obtain a complete knock-out mutant in the lines tested so far.
