Study of the biogenesis of monovalent-cation transporters in eukaryotic cell

dc.description.abstractRegulace vnitrobuněčné hladiny kationtů alkalických kovů (Na+ , K+ ) a protonů v buňkách většiny organismů (od bakterií po savce) závisí na správné lokalizaci a fungování transportérů těchto kationtů v membránách buněk. Cargo receptory jako součást endoplazmatického retikula a váčků COPII rozpoznávají membránové proteiny a usnadňují jim procházet skrze sekreční dráhu do jejich cílových membrán. Rodina cargo receptorů Erv14/Cornichon je konzervovaná napříč eukaryotními organismy a její členové se podílejí na biogenezi několika transportérů monovalentních katiotů. V této diplomové práci jsme podrobně analyzovali strukturu a funkce vybraných cornichonů z kvasinky Saccharomyces cerevisiae (Erv14) a z člověka (CNIH1-4). Pomocí heterologní exprese lidských cargo receptorů CNIH1, CNIH2 a CNIH4 v kvasince S. cerevisiae jsme zjištovali jejich schopnost nahradit úlohu cargo receptoru Erv14 v toleranci buněk k solím, udržování membránového potenciálu a vnitrobuněčného pH. Ze tří lidských cornichonů byly funkce Erv14 v udržování homeostáze monovalentních kationtů nejlépe komplementovány izoformou CNIH1. Exprese proteinů CNIH v S. cerevisiae také ovlivňovala biogenezi a funkci kvasinkového Na+ , K+ /H+ antiporteru Nha1. dc.description.abstractRegulation of intracellular levels of alkali metal cations (Na+ , K+ ) and protons depends on the proper localization and function of monovalent-cation transporters in cell membranes of most organisms (from bacteria to mammals). Cargo receptors, as a part of the endoplasmic reticulum and COPII vesicles, recognize membrane proteins and help them pass through the secretory pathway to reach their target membranes. The family of cargo receptors Erv14/Cornichon is conserved among eukaryotic organisms, and particular members participate in the biogenesis of several monovalent-cation transporters. In this diploma thesis, we analysed the structure and functions of selected cornichons from the yeast Saccharomyces cerevisiae (Erv14) and from humans (CNIH1-4) in more detail. Using a heterologous expression of human cargo receptors CNIH1, CNIH2, and CNIH4 in yeast S. cerevisiae, we studied their ability to fulfil the role of Erv14 cargo receptor in cell salt tolerance, maintenance of membrane potential and intracellular pH. Of the three human cornichons, the functions of Erv14 in maintaining monovalent cation homeostasis were best complemented by the CNIH1 isoform.
