dc.description.abstract5'-adenosinmonofosfátem aktivovaná proteinová kinasa je významným regulátorem buněčného metabolismu, růstu, proliferace, ale i například autofagie. Dle posledních výzkumů hraje důležitou roli i v patogenezi nervosvalového onemocnění myotonická dystrofie typu 1 a je možným terapeutickým cílem. V této práci byly kultivovány diferencované a nediferencované lidské svalové buněčné linie zdravých jedinců a pacientů s myotonickou dystrofií typu 1. Předmětem práce je studium a porovnání hladin AMPK v kulturách zdravých a nemocných buněk. Dalším bodem práce je afinitní purifikace tohoto enzymu metodou imunoprecipitace pomocí magnetických kuliček s navázanou protilátkou a následně hmotnostně spektrometrická analýza purifikovaného vzorku. Jelikož se spolu s AMPK podařilo afinitně purifikovat i další proteiny její signalizační kaskády, je část práce věnována také popisu těchto interakcí a porovnání množství koimunoprecipitovaných proteinů ve zdravých a nemocných buňkách. dc.description.abstract5' adenosine monophosphate-activated protein kinase is an important regulator of cell metabolism, growth, proliferation, as well as autophagy. According to latest research, AMPK plays an important role in the pathogenesis of the neuromuscular disorder myotonic dystrophy type 1 and is a possible therapeutic target. In this thesis, differentiated and proliferating human muscle cell lines from healthy individuals and myotonic dystrophy type 1 patients were cultivated. The thesis is focused on the study and comparison of AMPK levels in the cultures and affinity purification of AMPK using the method of immunoprecipitation with magnetic beads containing a bound antibody. The purified enzyme was analysed using mass spectrometry. Along with AMPK, parts of its signalling cascade were purified, a section of the thesis is therefore dedicated to the description of AMPK's interaction network and to the comparison of the levels of coimmunoprecipied proteins in cells from healthy individuals and myotonic dystrophy type 1 patients.
