Na střídavé resorpci a novotvorbě mají hlavní podíl osteoklasty, osteoblasty a osteocyty. Osteoklasty jsou jediné známé buňky se schopností odbourávat kostní tkáň. Tyto buňky vznikají fúzi prekurzorů v kostní dřeni nebo monocytů v periferní krvi v procesu zvaném osteoklastogeneze. Vznik osteoklastů z periferních prekurzorů může hrát významnou roli v souvislosti s chronickými zánětlivými chorobami, u nichž je známé zvýšené riziko rozvoje osteoporózy v průběhu života. Jednou z těchto chorob je celiakie, při níž u postižených jedinců po příjmu lepku v potravě dochází k rozpadu střevní sliznice a rozvoji chronického zánětu. V této práci jsme se zaměřili na potenciální roli osteoklastogeneze při rozvoji osteoporózy u pacientů s celiakií a ověřili jsme obecně vlivy vybraných zánětlivých agens (TNF-α, IL-6, IFN-γ a cfDNA) s předpokládanými nebo hypotetizovanými účinky v osteoklastogenezi. Materiál a metody: V plazmě a séru nově diagnostikovaných pacientů s celiakií, pacientů na dietě a zdravých kontrol jsme analyzovali koncentrace cfDNA a cytokinů TNF-α, IL-6, IFN- γ. Séra těchto pacientů a kontrol jsme spolu s rekombinantními cytokiny užívali v kultivacích. Pro ověření vlivu cfDNA jsme séra ošetřovali DNázou....
Introduction: Bone is a highly active tissue throughout life and is a subject to constant remodelling. Main cells responsible for continuous resorption and de novo synthesis of bone matrix are osteoclast, osteoblasts and osteocytes. Osteoclasts are the only known type of cells able to resorb bone. These cells are formed by fusion of precursor cells in bone marrow or peripheral blood in a process called osteoclastogenesis. Formation of osteoclasts may be of importance concerning chronic inflammatory diseases that are linked with higher risk of developing osteoporosis during lifespan. Celiac disease is one of those diseases, which is characterized by destruction of intestinal mucosa after ingestion of gluten by susceptible individuals followed by induction of chronic inflammation. In this work, we focused on the potential role of osteoclastogenesis in the development of osteoporosis in patients with celiac disease and we studied roles of selected inflammatory agents (TNF-α, IL-6, IFN-γ a cfDNA) with supposed or hypothesised effects on osteoclastogenesis.
