This dissertational thesis focuses on the clinical anatomy of the pubic symphysis, aiming to establish a comprehensive anatomical and functional model for pelvic ring surgery. Given the high failure rate of current surgical fixations in isolated symphyseolyses, a multimodal approach was adopted, combining a retrospective analysis of 652 patients' imaging data, a prospective dissectional study of 21 body donors' specimens, and numerical modelling using the finite element method. Morphometric results revealed significant individual variability and revised standard dimensions of the interpubic disc, which reaches a height of up to 51 mm in males. Dissectional analysis confirmed the almost constant presence of the anterior pubic ligament, which is absent from official anatomical nomenclature. In patients after birth-related trauma, a significant thickening of the ligamentous apparatus was identified as a result of permanent functional remodelling. Biomechanical simulations identified the ventro-caudal segment of the interpubic disc as the zone of maximum stress, correlating with the localisation of the interpubic cavity and supporting the hypothesis of its adaptive origin. The study concludes that the current standard of rigid plate osteosynthesis for isolated symphyseolysis is biomechanically...; Tato dizertační práce se zabývá klinickou anatomií stydké spony s cílem vytvořit komplexní anatomický a funkční model této struktury pro potřeby chirurgie pánevního kruhu. Vzhledem k vysoké incidenci selhání stávajících operačních fixací u izolovaných symfyzeolýz byl zvolen multimodální přístup kombinující retrospektivní analýzu obrazové dokumentace souboru 652 pacientů, prospektivní pitevní studii 21 preparátů těl dárců a numerické modelování metodou konečných prvků. Výsledky morfometrické části prokázaly výraznou individuální variabilitu a revidovaly učebnicové rozměry interpubického disku, který u mužů dosahuje výšky až 51 mm. Pitevní analýza potvrdila takřka konstantní přítomnost předního stydkého vazu, jenž není součástí oficiální anatomické nomenklatury. U pacientek po prodělaném porodním traumatu bylo zjištěno signifikantní zesílení vazivového aparátu v důsledku trvalé funkční remodelace. Biomechanická simulace identifikovala ventrokaudální segment interpubického disku jako zónu maximálního napětí, což koreluje s umístěním interpubické dutiny a podporuje hypotézu o jejím adaptivním vzniku. Cognitive Brain Mapping with Intracranial EEG ABSTRACT Mapping cognitive functions is a central topic in neuroscience, aiming to localize cognitive processes within specific brain structures and functional networks and describing their spatiotemporal dynamics. Intracranial electroencephalography (iEEG), as an invasive technique, offers a unique combination of direct measurement of neuronal activity within the brain with high temporal resolution and spatial precision. This thesis focuses on elucidating neuronal dynamics, functional connectivity, and interregional interactions during the performance of selected cognitive tasks. The research is structured into three thematic areas: 1) the study of numerical cognition with a focus on elementary arithmetic operations, 2) mechanisms of attention switching between internally and externally directed attention, and 3) interactions between the ventral and dorsal visual streams during memory-guided actions. The analyses include iEEG data collected from 38 patients with drug-resistant epilepsy between 2020 and 2024. A unique experimental paradigm was developed for each thematic area. The iEEG signal was processed using software packages in MATLAB, and advanced methods of time-frequency analysis and functional connectivity were applied to characterize events at the...; 1 Mapování kognitivních funkcí pomocí intrakraniálního EEG ABSTRAKT Mapování kognitivních funkcí je stěžejním tématem v neurovědách, jehož cílem je lokalizovat kognitivní procesy v rámci anatomických a funkčních oblastí mozku a popsat jejich časoprostorovou dynamiku. Intrakraniální elektroencefalografie (iEEG) nabízí jako invazivní metoda unikátní kombinaci vysokého časového rozlišení, prostorové specifity a přímého měření neuronální aktivity z mozku. Předkládaná disertační práce se zaměřuje na objasnění neuronální dynamiky, funkční konektivity a interregionálních interakcí během řešení vybraných kognitivních úloh. Výzkum je strukturován do tří tematických okruhů: 1) studium numerické kognice se zaměřením na elementární aritmetické operace, 2) mechanismy přepínání pozornosti mezi interně a externě orientovanými podněty, a 3) interakce ventrálního a dorzálního vizuálního proudu během akcí, které jsou řízeny pamětí. Analýza zahrnovala iEEG data získaná od celkem 38 pacientů s farmakorezistentní epilepsií v letech 2020-2024. Pro každý okruh bylo vyvinuto unikátní experimentální paradigma. <summary type="text">Membránové vlastnosti neurálních kmenových/progenitorových buněk v průběhu in vitro diferenciace a po transplantaci do mozku potkana
The aim ofthis work was to elucidate the differentiation mechanisms ofneural stem/progenitor cells in vitro and their potential to survive and differentiate in vivo, after transplantation into the injured rat cortex. Immunohistochemistry was used for cell identifications, and the properties ofK+and Na+ voltage-gated ion channels were studied using the patch-clamp technique. We have demonstrated that immortalised green fluorescent protein (GFP)/NE-4C neural stem cells derived from the neuroepithelium of p53"deficient mouse embryos at embryonic day (E)9 are able to differentiate into neurons in vitro. After transplantation into the site ofa photochemical lesion ofadult 'rats, GFP/NE-4C cells survive and give rise to neurons, astrocytes and oligodendrocytes. Primary embryonic neural stem cells were isolated from D6/GFP mice, in which GFP is expressed under the control ofD6, a promoter of the mDachl gene, which is involved in the development of the cortex. At El2, D6 is specifically expressed in the neural stem cells of the dorsal telencephalon, from which cortical neurons arise. We have shown that D6/GFP neural stem cells isolated from E12 embryos are able to give rise to neurons and glial fibrillary acidic protein (GFAP)-positive cells in vitro and that after transplantation intó the non-injured...; Cílem této práce bylo objasnění mechanismů diferenciace neurálních kmenových/progenitorových buněk in vitro a jejich přežívání a diferenciace po transplantaci do poškozené kůry mozku potkana. Pro identifikaci buněk byla použita imunohistochemická analýza a změny v expresi Na+ a K+ napěťově závislých iontových kanálů, a receptorů byly sledovány pomocí metody patch-clamp. Ukázali jsme, že NE-4C imortilizované neurální kmenové buňky, které byly získány z neuroepitelia p53-deficientních myších embryí devátý embryonální den (E9) a označeny zeleným fluorescenčním proteinem (GFP/NE-4C), mají schopnost diferenciace v neurony in vitro. Po transplantaci do fotochemické léze dávají tyto buňky vznik neuronům, astrocytům i oligodendrocytům. Dále byly použity primární embryonální kmenové buňky izolované z embryí D6/GFP myší, které exprimují GFP pod D6 promotorem mDachl genu, který se uplatňuje při vývoji mozkové kůry a je specificky exprimován v neurálních kmenových buňkách mozkové kt1ry a ve vzniklých neuronech. V El2 obsahuje mozková kůra u těchto myší převážně proliferující neurální kmenové buňky, které je tak možno na základě fluorescence GFP izolovat.
